In the MAN Lion coach there is a button (via cockpit view 7 or), trailing axle relief. I am wondering what this does if you turn it on ... as well what it does if it is off.

I did not check this carefully because I have a more automatic driving setting, maybe a stop brake, test and you'll see. @pete_agreatguy

Guessing by the name it is supposed to lift/drop the 3rd axle on tri-axle buses, e.g. Lion's Coach C. Though there are some buttons without actual functions.
Nope, it's not lift/drop the rear axle.

It is normally turned off. But when it is turned on for example in slippery conditions more weight is on drive axle and therefore more friction which helps to accelerate the bus from standstill. It is used at low speeds only. It has no real function in this game to my knowledge.
